Prolog wyścigu został rozegrany po ulicach miasta Luksemburg, przy sztucznym świetle. Ostatni zawodnik, Borut Bozic (Vacansoleil-DCM), na trasę wyruszył o 21:12.
Zawodnicy do pokonania mieli 2,6 kilometra, po krętych uliczkach. Najlepiej z przeszkodami, poradził sobie Szwajcar Fabian Cancellara. Mistrz Świata w jeździe na czas, wykręcił rezultat 3:42. Pięć sekund do niego stracił torowiec, Damien Gaudin. Trzeci, doświadczony Engoulvent, do zwycięzcy stracił 8 sekund.
Polacy Maciej Bodnar (Liquigas-Cannondale) oraz Michał Kwiatkowski (RadioShack), choć z czasówkami są za pan brat, to dystans był dla nich nieco za krótki. „Bodi” do zwycięzcy stracił 28 sekund, zajmując 80. miejsce. Z kolei „Kwiatek”, do Cancellary miał 31 sekund, wykręcając 104. czas.
